Die vorliegende Masterarbeit belegt, dass ein Innovationsvorhaben mittels „Bottom-up“-Management
zur Entwicklung einer gesamten Organisation führen kann. Dies wird exemplarisch an
der Curriculumentwicklung an einer Pflegeschule in Baden-Württemberg dargestellt. Hierfür
werden zunächst die Besonderheiten des „Bottom-up“-Managements herausgearbeitet und
anschließend unter Einbeziehung aller Rahmenbedingungen ein eigenes Handlungskonzept
für das Projektvorhaben erstellt. Das „Drei-Wege-Modell der Schulentwicklung“ von Rolff, welches
sich mit dem Zusammenhang zwischen der Unterrichtsentwicklung, der Personalentwicklung
und der Organisationsentwicklung befasst (vgl. Rolff 2016, S. 20), stellt in dieser
Arbeit neben der Qualitätssicherung einen wichtigen Orientierungsrahmen dar. Es wird zudem
gezeigt, dass sowohl das Führungsverhalten der Schulleitung als auch die Möglichkeit
zur partizipativen Mitarbeit aller Teilnehmenden der Schlüssel für das Gelingen des Innovationsvorhabens
ist. Damit die Ergebnisse dieser Arbeit auch gewinnbringend für andere Projektvorhaben
und Organisationen genutzt werden können, werden sie abschließend auf ihre
Übertragbarkeit hin geprüft. So können Teilaspekte diese Masterarbeit auch bei der Weiterentwicklung
weiterer Pflegeschulen oder anderer Organisationen eine Hilfe sein.
Das Kyoto-Protokoll, welches die letzten anderthalb Dekaden für Industrieländer erstmalig völkerrechtlich bindende Emissionsreduktionsziele für Treibhausgase festschrieb, läuft in diesem Jahr aus. Die im internationalen Abkommen entwickelten Klimaschutzin-strumente in Form eines Handels mit Emissionszertifikaten, sowie in Form von Emissi-onsreduktionen mittels Klimaschutzprojekten in Drittländern (Joint Implementation und Clean Development Mechanismus), konnten in der Vergangenheit nicht zum angestrebten Rückgang des weltweiten Treibhausgasausstoßes führen.
Das 2015 beschlossene Klimaabkommen von Paris soll die Regelungen des Kyoto-Protokolls nun ablösen und neue ambitionierte Standards im Klimaschutz setzen, um die Klimaerwärmung langfristig auf maximal 2 °C gegenüber des vorindustriellen Ni-veaus zu beschränken. Die grundlegend veränderten Rahmenbedingungen des Pariser Vertrags, sowie die Erfahrungen mit bisherigen flexiblen Instrumenten der Klimarah-menkonvention der Vereinten Nationen, erschweren aufgrund der hohen Komplexität und teils divergierender Interessenlagen der beteiligten Akteure seit 2015 sowohl die Entwicklung als auch die internationale Einigung auf einen neuen Marktmechanismus.
Die vorliegende Arbeit untersucht auf verschiedenen Ebenen, welche Vor- und Nachteile sich aus dem CDM ergeben haben und wie diese in die Ausgestaltung von Artikel 6 des Pariser Klimaschutzabkommens miteinfließen können. Dabei werden auch die Themen Klimakompensation und Klimaneutralität behandelt, welche zuletzt einen enormen Bedeutungszuwachs verzeichnen konnten und im Privatsektor inzwischen ein eminentes Nachfragepotential bergen. Im Zusammenhang mit den Attraktivitätsfaktoren einer Investition in Projekte, die nachhaltige Entwicklung fördern, werden hierzu auch die möglichen Hintergründe des verstärkten Engagements seitens der Unternehmen diskutiert.
Das theoretische Konzept der Arbeit basiert auf dem Free Rider Problem und den öko-logischen Managementregeln. Der Fokus liegt auf ökologischen Aspekten der Nachhaltigkeit, wobei ebenso ökonomische Komponenten ausschlaggebend sind.
Als ergänzendes Klimaschutzinstrument wird der Handel mit Emissionszertifikaten aufgegriffen, da er in der EU bereits etabliert ist.
Eine der größten Herausforderungen scheint es, wirtschaftliches Wachstum und Klimaschutz zu vereinen und Entwicklungsländern den Weg für eine Industrialisierung bzw. anziehende wirtschaftliche Entwicklung zu ebnen, welche keine nachholenden Ansprüche auf Emissionen stellt, sondern sich innovativ und inklusiv gestaltet. Für die Überwindung dieser derzeit bestehenden Kontroverse, die in engem Zu-sammenhang mit der Diskussion um einen neuen umfassenden Klimaschutzmechanismus steht, werden schließlich Empfehlungen ausgesprochen und ein abschließender Ausblick – auch unter Berücksichtigung der aktuellen Klimaschutzentwicklungen in Fol-ge der weltweiten COVID-19-Pandemie gewährt.
Riesling wird neben seiner mannigfaltigen Variabilität im Aromaprofil, das unter anderem durch die unterschiedliche Bodenbeschaffenheit entsteht, vor allem in Deutschland auch wegen seiner Kältetoleranz und Anpassungsfähigkeit geschätzt. Er gilt zudem auch als alterungsfähiger Wein, allerdings kommt es bei zu starker Sonnenexposition der Rebe und langer bzw. warmer Lagerung vermehrt zur Bildung von 1,1,6-Trimethyl-1,2-dihydronaphthalin (TDN). Dieser von Carotinoiden abstammende Aromastoff verursacht die sogenannte „Petrolnote“, die vor allem in wärmeren Anbauregionen zum sortentypischen Bukett des Rieslings gehört. Deutsche Rieslingweine zeichnen sich dagegen überwiegend durch einen säurebetonten, fruchtigen Charakter aus, bei dem das Auftreten einer Petrolnote vor allem im Fall von Jungweinen als unpassende Fehlnote empfunden wird.
Das Ziel der vorliegenden Arbeit war deswegen, die sensorische Relevanz von TDN zu evaluieren und Maßnahmen zu realisieren, die geeignet sind, die Konzentrationen an freiem und gebundenem TDN zu verringern und dadurch das Auftreten der Petrolnote zu vermeiden.
Dafür wurde zunächst in Kapitel 6.1 die Empfindlichkeit von Verbrauchern und geschulten Prüfern gegenüber TDN sowie die Konzentration bestimmt, ab der die Petrolnote zu einer Ablehnung des Weins durch Verbraucher führt. Während geschulte Prüfer Rieslingweine bereits ab einem TDN-Gehalt von 2,3 µg/L unterscheiden konnten, lag die Wahrnehmungsschwelle von 156 Verbrauchern mit 14,7 µg/L um ein Mehrfaches darüber, und wurde außerdem durch das Geschlecht der Probanden beeinflusst. Die Petrolnote führte ab TDN-Gehalten von 60 µg/L bei einjährigem und 91 µg/L bei achtjährigem Riesling zur Ablehnung des Weins. Die Konzentration an freiem TDN in 261 Rieslingweinen aus drei verschiedenen Weinwettbewerben überstieg bei rund der Hälfte der Weine die Wahrnehmungsschwelle von geschulten Prüfern, während die Wahrnehmungsschwelle von Verbrauchern nur von 15% der Weine überschritten wurde. Gleichzeitig lag bei keinem der Weine der TDN-Gehalt über der Ablehnungsschwelle.
Durch die Evaluierung der instrumentellen Analyseparameter in Kapitel 6.2 wurde für die Untersuchung von freiem TDN und weiteren Aromastoffen eine Methode entwickelt, die es ermöglicht, nicht nur die TDN-Konzentrationen zu erfassen, sondern auch eine umfassende Qualitätsbewertung der Versuchsweine durchzuführen. Parallel dazu wurde eine Schnellmethode zur Erfassung der Gehalte an gebundenem TDN und Vitispiran implementiert, um auch die Effektivität der in dieser Arbeit durchgeführten weinbaulichen und oenologischen Praktiken im Hinblick auf das TDN-Potential zu beurteilen.
Kapitel 6.3 und 6.4 beschreiben weinbauliche Maßnahmen, die in mehrjährigen Studien auf ihre Eignung zur Reduzierung der TDN-Konzentration untersucht wurden. Während bei den Weinen, die aus Beeren unterschiedlicher Größe hergestellt wurden, keine signifikanten Unterschiede über die Jahrgänge hinweg beobachtet wurden, konnte durch die Variation der Rebunterlagen der Gehalt an gebundenem TDN um rund 30% reduziert werden. Ausgangspunkt einer weiteren Versuchsreihe waren acht verschiedene Rieslingklone auf derselben Unterlage, welche anschließend auf ihren TDN Gehalt untersucht wurden. Dabei wurden deutliche Differenzen in der Disposition einiger Klone zu höheren Gehalten an gebundenem TDN festgestellt. Hier ergab sich eine positive Korrelation zwischen der Lockerbeerigkeit der Trauben und der Menge an gebundenem TDN in den produzierten Weinen – je kompakter die Traube, desto weniger gebundenes TDN und gebundenes Vitispiran wurde gebildet. Die höhere Sonnenexposition der Beeren, die diesen Effekt hervorruft, beeinflusste auch die Gehalte an gebundenem TDN und Vitispiran in Weinen, die von Reben geerntet wurden, welche zu unterschiedlichen Zeitpunkten und in variierender Intensität entblättert wurden. Dabei führt sowohl eine maximale Entblätterung in der Traubenzone wie auch die Laubentfernung einen Monat nach der Blüte zu einer Erhöhung der Konzentration an gebundenem TDN und Vitispiran von rund 50%. Entblätterungsmaßnahmen zur Blüte oder zur Véraison, die der Regulierung der Erntemenge und der Traubengesundheit dienen, führten dagegen zu keinem Anstieg im Vergleich zur nicht-entblätterten Kontrolle.
Wie in Kapitel 6.5 ausgeführt wird, resultiert ein hoher Pressdruck beim Keltern sowie ein niedriger Stickstoffgehalt des Mosts in einer Zunahme des gebundenen TDN von 50 100%. Höhere Säuregehalte während der Lagerung verursachten in mehreren Versuchsreihen nicht nur eine höhere Freisetzungsrate von TDN, sondern auch einen verstärkten Abbau anderer Aromastoffe wie Ester, β-Damascenon oder Linalool. Dagegen hatte ein niedriger pH-Wert während der Gärung kaum Einfluss auf den Hefemetabolismus und die dadurch gebildeten Aromastoffe. Die Erhöhung der Gärtemperatur von 12 auf 24 °C hatte jedoch eine Zunahme von honig- oder petrolartigen Noten in den Rieslingweinen zur Folge. Die Verwendung unterschiedlicher Hefestämme führte zu einer Variation der Konzentrationen an gebundenem TDN zwischen 70 und 147 µg/L, abhängig vom Hefestamm und dem Jahrgang. Zwei der untersuchten neun Hefen brachten Weine mit bis zu 40% geringeren Gehalten an gebundenem TDN in Mosten mit hohem Stickstoffgehalt hervor, während drei weitere Hefen besser für den Einsatz in nährstoffarmen Most geeignet waren. Bei der Lagerung der Weine spielte die Lagertemperatur eine entscheidende Rolle in Bezug auf den Gehalt an freiem TDN, gefolgt vom Material des Flaschenverschlusses und der Flaschenorientierung.
Mittels geeigneter Filtermaterialien, die in Kapitel 7 beschrieben sind, wurde der Gehalt an freiem Wein um bis zu 80% reduziert, ohne die meisten der anderen Aromastoffe signifikant zu beeinflussen.
Somit wurde durch diese Arbeit ein vielfältiger Maßnahmenkatalog für die Weinwirtschaft entwickelt, der geeignet ist, den Anforderungen des fortschreitenden Klimawandels entgegenzutreten und die herausragende Position des Rieslings in Deutschland zu sichern.
The plasma membrane transporter SOS1 (SALT-OVERLY SENSITIVE1) is vital for plant survival under salt stress. SOS1 activity is tightly regulated, but little is known about the underlying mechanism. SOS1 contains a cytosolic, autoinhibitory C-terminal tail (abbreviated as SOS1 C-term), which is targeted by the protein kinase SOS2 to trigger its transport activity. Here, to identify additional binding proteins that regulate SOS1 activity, we synthesized the SOS1 C-term domain and used it as bait to probe Arabidopsis thaliana cell extracts. Several 14-3-3 proteins, which function in plant salt tolerance, specifically bound to and interacted with the SOS1 C-term. Compared to wild-type plants, when exposed to salt stress, Arabidopsis plants overexpressing SOS1 C-term showed improved salt tolerance, significantly reduced Na+ accumulation in leaves, reduced induction of the salt-responsive gene WRKY25, decreased soluble sugar, starch, and proline levels, less impaired inflorescence formation and increased biomass. It appears that overexpressing SOS1 C-term leads to the sequestration of inhibitory 14-3-3 proteins, allowing SOS1 to be more readily activated and leading to increased salt tolerance. We propose that the SOS1 C-term binds to previously unknown proteins such as 14-3-3 isoforms, thereby regulating salt tolerance. This finding uncovers another regulatory layer of the plant salt tolerance program
A branch-and-cut approach and alternative formulations for thetraveling salesman problem with drone

In this paper, we are interested in studying thetraveling salesman problem withdrone(TSP-D). Given a set of customers and a truck that is equipped with a singledrone, the TSP-D asks that all customers are served exactly once and minimal deliv-ery time is achieved. We provide two compact mixed integer linear programmingformulations that can be used to address instances with up to 10 customer within afew seconds. Notably, we introduce a third formulation for the TSP-D with an expo-nential number of constraints. The latter formulation is suitable to be solved by abranch-and-cut algorithm. Indeed, this approach can be used to find optimal solu-tions for several instances with up to 20 customers within 1 hour, thus challenging thecurrent state-of-the-art in solving the TSP-D. A detailed numerical study providesan in-depth comparison on the effectiveness of the proposed formulations. More-over, we reveal further details on the operational characteristics of a drone-assisteddelivery system. By using three different sets of benchmark instances, considera-tion is given to various assumptions that affect, for example, technological droneparameters and the impact of distance metrics.
A building-block model reveals new insights into the biogenesis of yeast mitochondrial ribosomes

Most of the mitochondrial proteins in yeast are encoded in the nuclear genome, get synthesized by cytosolic ribosomes and are imported via TOM and TIM23 into the matrix or other subcompartments of mitochondria. The mitochondrial DNA in yeast however also encodes a small set of 8 proteins from which most are hydrophobic membrane proteins and build core components of the OXPHOS complexes. They get synthesized by mitochondrial ribosomes which are descendants of bacterial ribosomes and still have some similarities to them. On the other hand, mitochondrial ribosomes experienced various structural and functional changes during evolution that specialized them for the synthesis of the mitochondrial encoded membrane proteins. The mitoribosome contains mitochondria-specific ribosomal proteins and replaced the bacterial 5S rRNA by mitochondria-specific proteins and rRNA extensions. Furthermore, the mitoribosome is tethered to the inner mitochondrial membrane to facilitate a co-translational insertion of newly synthesized proteins. Thus, also the assembly process of mitoribosomes differs from that of bacteria and is to date not well understood.
Therefore, the biogenesis of mitochondrial ribosomes in yeast should be investigated. To this end, a strain was generated in which the gene of the mitochondrial RNA-polymerase RPO41 is under control of an inducible GAL10-promoter. Since the scaffold of ribosomes is built by ribosomal RNAs, the depletion of the RNA-polymerase subsequently leads to a loss of mitochondrial ribosomes. Reinduction of Rpo41 initiates the assembly of new mitoribosomes, which makes this strain an attractive model to study mitoribosome biogenesis.
Initially, the effects of Rpo41 depletion on cellular and mitochondrial physiology was investigated. Upon Rpo41 depletion, growth on respiratory glycerol medium was inhibited. Furthermore, mitochondrial ribosomal 21S and 15S rRNA was diminished and mitochondrial translation was almost completely absent. Also, mitochondrial DNA was strongly reduced due to the fact that mtDNA replication requires RNA primers that get synthesized by Rpo41.
Next, the effect of reinduction of Rpo41 on mitochondria was tested. Time course experiments showed that mitochondrial translation can partially recover from 48h Rpo41 depletion within a timeframe of 4.5h. Sucrose gradient sedimentation experiments further showed that the mitoribosomal constitution was comparable to wildtype control samples during the time course of 4.5h of reinduction, suggesting that the ribosome assembly is not fundamentally altered in Gal-Rpo41 mitochondria. In addition, the depletion time was found to be critical for recovery of mitochondrial translation and mitochondrial RNA levels. It was observed that after 36h of Rpo41 depletion, the rRNA levels and mitochondrial translation recovered to almost 100%, but only within a time course of 10h.
Finally, mitochondria from Gal-Rpo41 cells isolated after different timepoints of reinduction were used to perform complexome profiling and the assembly of mitochondrial protein complexes was investigated. First, the steady state conditions and the assembly process of mitochondrial respiratory chain complexes were monitored. The individual respiratory chain complexes and the super-complexes of complex III, complex IV and complex V were observed. Furthermore, it was seen that they recovered from Rpo41 depletion within 4.5h of reinduction. Complexome profiles of the mitoribosomal small and large subunit discovered subcomplexes of mitoribosomal proteins that were assumed to form prior to their incorporation into assembly intermediates. The complexome profiles after reinduction indeed showed the formation of these subcomplexes before formation of the fully assembled subunit. In the mitochondrial LSU one subcomplex builds the membrane facing protuberance and a second subcomplex forms the central protuberance. In contrast to the preassembled subcomplexes, proteins that were involved in early assembly steps were exclusively found in the fully assembled subunit. Proteins that assemble at the periphery of the mitoribosome during intermediate and late assembly steps where found in soluble form suggesting a pool of unassembled proteins that supply assembly intermediates with proteins.
Taken together, the findings of this thesis suggest a so far unknow building-block model for mitoribosome assembly in which characteristic structures of the yeast mitochondrial ribosome form preassembled subcomplexes prior to their incorporation into the mitoribosome.
In grinding, the crystal grain size of the workpiece material is relatively same range compared to the removal depth. This raises a question if an anisotropic material model, which considers the effect of the crystal grain size and orientations, would better predict the process forces when compared to an isotropic material model. Initially, a simple micro-indentation process is chosen to compare the two models. In this work, a crystal plasticity model and an isotropic Johnson-Cooke plasticity model are employed to simulate micro-identation of a twinning induced plasticity (TWIP) steel. The results of the two models are compared using the force-displacement curves from the micro-indentation experiments. In the future, the study will be extended to describe the material removal process during a single grit scratch test.
A detailed study of a cylinder activation concept by efficiency loss analysis and 1D simulation

Cylinder deactivation is a well-known measure for reducing fuel consumption, especially when applied to gasoline engines. Mostly, such systems are designed to deactivate half of the number of cylinders of the engine. In this study, a new concept is investigated for deactivating only one out of four cylinders of a commercial vehicle diesel engine (“3/4-cylinder concept”). For this purpose, cylinders 2–4 of the engine are operated in “real” 3-cylinder mode, thus with the firing order and ignition distance of a regular 3-cylinder engine, while the first cylinder is only activated near full load, running in parallel to the fourth cylinder. This concept was integrated into a test engine and evaluated on an engine test bench. As the investigations revealed significant improvements for the low-to-medium load region as well as disadvantages for high load, an extensive numerical analysis was carried out based on the experimental results. This included both 1D simulation runs and a detailed cylinder-specific efficiency loss analysis. Based on the results of this analysis, further steps for optimizing the concept were derived and studied by numerical calculations. As a result, it can be concluded that the 3/4-cylinder concept may provide significant improvements of real-world fuel economy when integrated as a drive unit into a tractor.
We propose a model for glioma patterns in a microlocal tumor environment under
the influence of acidity, angiogenesis, and tissue anisotropy. The bottom-up model deduction
eventually leads to a system of reaction–diffusion–taxis equations for glioma and endothelial cell
population densities, of which the former infers flux limitation both in the self-diffusion and taxis
terms. The model extends a recently introduced (Kumar, Li and Surulescu, 2020) description of
glioma pseudopalisade formation with the aim of studying the effect of hypoxia-induced tumor
vascularization on the establishment and maintenance of these histological patterns which are typical
for high-grade brain cancer. Numerical simulations of the population level dynamics are performed
to investigate several model scenarios containing this and further effects.
Various regulatory initiatives (such as the pan-European PRIIP-regulation or the German chance-risk classification for state subsidized pension products) have been introduced that require product providers to assess and disclose the risk-return profile of their issued products by means of a key information document. We will in this context outline a concept for a (forward-looking) simulation-based approach and highlight its application and advantages. For reasons of comparison, we further illustrate the performance of approximation methods based on a projection of observed returns into the future such as the Cornish–Fisher expansion or bootstrap methods.
A highly water-dispersible heterogeneous Brønsted acid surfactant was prepared by synthesis of a bi-functional anisotropic Janus-type material. The catalyst comprises ionic functionalities on one side and propyl-SO3H groups on the other. The novel material was investigated as a green substitute of a homogeneous acidic phase transfer catalyst (PTC). The activity of the catalyst was investigated for the aqueous-phase oxidation of cyclohexene to adipic acid with 30 % hydrogen peroxide even in a decagram-scale. It can also be used for the synthesis of some other carboxylic acid derivatives as well as diethyl phthalate.
We have investigated urine samples after coffee consumption using targeted and untargeted
approaches to identify furan and 2-methylfuran metabolites in urine samples by UPLC-qToF.
The aim was to establish a fast, robust, and time-saving method involving ultra-performance
liquid chromatography-quantitative time-of-flight tandem mass spectrometry (UPLC-qToF-MS/MS).
The developed method detected previously reported metabolites, such as Lys-BDA, and others that
had not been previously identified, or only detected in animal or in vitro studies. The developed
UPLC-qToF method detected previously reported metabolites, such as lysine-cis-2-butene-1,4-dial
(Lys-BDA) adducts, and others that had not been previously identified, or only detected in animal
and in vitro studies. In sum, the UPLC-qToF approach provides additional information that may be
valuable in future human or animal intervention studies.
Solar radiation data is essential for the development of many solar energy applications ranging from thermal collectors to building simulation tools, but its availability is limited, especially the diffuse radiation component. There are several studies aimed at predicting this value, but very few studies cover the generalizability of such models on varying climates. Our study investigates how well these models generalize and also show how to enhance their generalizability on different climates. Since machine learning approaches are known to generalize well, we apply them to truly understand how well they perform on different climates than they are originally trained. Therefore, we trained them on datasets from the U.S. and tested on several European climates. The machine learning model that is developed for U.S. climates not only showed low mean absolute error (MAE) of 23 W/m2, but also generalized very well on European climates with MAE in the range of 20 to 27 W/m2. Further investigation into the factors influencing the generalizability revealed that careful selection of the training data can improve the results significantly
A novel shadowgraphic inline probe to measure crystal size distributions (CSD),
based on acquired greyscale images, is evaluated in terms of elevated temperatures and fragile
crystals, and compared to well-established, alternative online and offline measurement techniques,
i.e., sieving analysis and online microscopy. Additionally, the operation limits, with respect to
temperature, supersaturation, suspension, and optical density, are investigated. Two different
substance systems, potassium dihydrogen phosphate (prisms) and thiamine hydrochloride (needles),
are crystallized for this purpose at 25 L scale. Crystal phases of the well-known KH2PO4/H2O system
are measured continuously by the inline probe and in a bypass by the online microscope during
cooling crystallizations. Both measurement techniques show similar results with respect to the crystal
size distribution, except for higher temperatures, where the bypass variant tends to fail due to
blockage. Thiamine hydrochloride, a substance forming long and fragile needles in aqueous solutions,
is solidified with an anti-solvent crystallization with ethanol. The novel inline probe could identify
a new field of application for image-based crystal size distribution measurements, with respect
to difficult particle shapes (needles) and elevated temperatures, which cannot be evaluated with
common techniques.
Phase field modeling of fracture has been in the focus of research for over a decade now. The field has gained attention properly due to its benefiting features for the numerical simulations even for complex crack problems. The framework was so far applied to quasi static and dynamic fracture for brittle as well as for ductile materials with isotropic and also with anisotropic fracture resistance. However, fracture due to cyclic mechanical fatigue, which is a very important phenomenon regarding a safe, durable and also economical design of structures, is considered only recently in terms of phase field modeling. While in first phase field models the material’s fracture toughness becomes degraded to simulate fatigue crack growth, we present an alternative method within this work, where the driving force for the fatigue mechanism increases due to cyclic loading. This new contribution is governed by the evolution of fatigue damage, which can be approximated by a linear law, namely the Miner’s rule, for damage accumulation. The proposed model is able to predict nucleation as well as growth of a fatigue crack. Furthermore, by an assessment of crack growth rates obtained from several numerical simulations by a conventional approach for the description of fatigue crack growth, it is shown that the presented model is able to predict realistic behavior.
The cultivation of cyanobacteria with the addition of an organic carbon source (meaning as heterotrophic or mixotrophic cultivation) is a promising technique to increase their slow growth rate. However, most cyanobacteria cultures are infected by non-separable heterotrophic bacteria. While their contribution to the biomass is rather insignificant in a phototrophic cultivation, problems may arise in heterotrophic and mixotrophic mode. Heterotrophic bacteria can potentially utilize carbohydrates quickly, thus preventing any benefit for the cyanobacteria. In order to estimate the advantage of the supplementation of a carbon source, it is essential to quantify the proportion of cyanobacteria and heterotrophic bacteria in the resulting biomass. In this work, the use of quantitative polymerase chain reaction (qPCR) is proposed. To prepare the samples, a DNA extraction method for cyanobacteria was improved to provide reproducible and robust results for the group of terrestrial cyanobacteria. Two pairs of primers were used, which bind either to the 16S rRNA gene of all cyanobacteria or all bacteria including cyanobacteria. This allows a determination of the proportion of cyanobacteria in the biomass. The method was established with the two terrestrial cyanobacteria Trichocoleus sociatus SAG 26.92 and Nostoc muscorum SAG B-1453-12a. As proof of concept, a heterotrophic cultivation with T. sociatus with glucose was performed. After 2 days of cultivation, a reduction of the biomass partition of the cyanobacterium to 90% was detected. Afterwards, the proportion increased again.
Personalized dynamic pricing (PDP) involves dynamically setting individual-consumer prices for the same product or service according to consumer-identifying information. Despite its profitability, this pricing provokes strong negative fairness perceptions, explaining why managers are reluctant to implement it. This research provides important insights into the effect of two PDP dimensions (price individualization level and segmentation base) on fairness perceptions and the moderating role of privacy concerns. The results of two experimental studies indicate that consumers perceive individual prices as less fair than segment prices. They also evaluate location-based pricing as less fair than purchase history-based pricing. Consumer privacy concerns moderate these effects.
One technique to describe the failure of mechanical structures is a phase field model for fracture. Phase field models for fracture consider an independent scalar field variable in addition to the mechanical displacement [1]. The phase field ansatz approximates crack surfaces as a continuous transition zone in which the phase field variable varies from a value that indicates intact material to another value that represents cracks. For a good approximation of cracks, these transition zones are required to be narrow, which leads to steep gradients in the fracture field. As a consequence, the required mesh density in a finite element simulation and thus the computational effort increases. In order to circumvent this efficiency problem, exponential shape functions were introduced in the discretization of the phase field variable, see [2]. Compared to the bilinear shape functions these special shape functions allow for a better approximation of the steep transition with less elements. Unfortunately, the exponential shape functions are not symmetric, which requires a certain orientation of elements relative to the crack surfaces. This adaptation is not uniquely determined and needs to be set up in the correct way in order to improve the approximation of smooth cracks. The issue is solved in this work by reorientating the exponential shape functions according to the nodal value of phase field gradient in a particular element. To be precise, this work discusses an adaptive algorithm that implements such a reorientation for 2d and 3d situations.
Machining is very common in industry, e.g. automotive industry and aerospace industry, which is a nonlinear dynamic problem including large deformations, large strain, large strain rates and high temperatures, that implies some difficulties for numerical methods such as Finite element method. One way to simulate such kind of problems is the Particle Finite Element Method (PFEM) which combines the advantages of continuum mechanics and discrete modeling techniques. In this work we introduce an improved PFEM called the Adaptive Particle Finite Element Method (A-PFEM). The A-PFEM introduces particles and removes wrong elements along the numerical simulation to improve accuracy, precision, decrease computing time and resolve the phenomena that take place in machining in multiple scales. At the end of this paper, some examples are present to show the performance of the A-PFEM.
Additive 3D-Drucksverfahren ermöglichen eine automatisierte wie flexible Fertigung
komplexer 3D-Geometrien direkt aus einem CAD-Modell ohne die Notwendigkeit ei
nes bauteilspezifischen Werkzeugs. Nachteil vor allem beim 3D-Drucken von Kunst
stoffen sind jedoch die geringen mechanischen Eigenschaften, die auf verfahrensbe
dingte Herausforderungen, aber auch auf eine eingeschränkte Auswahl verarbeitba
rer Materialien zurückzuführen sind. Eine Möglichkeit die mechanischen Eigenschaf
ten von Kunststoffen zu verbessern, ist die Kombination mit Verstärkungsfasern. Die
höchste Verstärkungswirkung entfalten Faser-Kunststoff-Verbunde (FKV) wenn die
Fasern kontinuierlich und in Lastrichtung vorliegen. Um ihr volles Potential zu entfal
ten, müssen FKV daher möglichst gut an die jeweiligen Anwendungen angepasst
werden. Das erschwert eine automatisierte und effiziente Fertigung, gerade von
komplexeren Strukturen. Ziel der Arbeit war daher die Entwicklung eines 3D
Verfahrens für kontinuierlich faserverstärkte Kunststoffe. Hierdurch soll das Anwen
dungsspektrum kunststoffbasierter 3D-Druck-Verfahren vergrößert und gleichzeitig
eine effiziente sowie flexible Fertigung komplexer FKV-Strukturen ermöglicht werden.
Das entwickelte Prozesskonzept basiert dabei auf 3D-Druck-Extrusionsverfahren für
thermoplastische Kunststoffe. Im sogenannten Fiber Integrated Fused Deposition
Modeling Prozess, kurz FIFDM, werden bereits imprägnierte Halbzeuge in Form von
kontinuierlich faserverstärkten Thermoplaststrängen (FTS) verarbeitet. Um die Fa
serorientierung frei einstellen zu können, werden die Stränge nicht wie herkömmlich
nur schichtweise, sondern frei in alle Raumrichtungen positioniert. Realisiert wird dies
über die Steuerung der FTS-Temperatur nach der Extrusion. Im Rahmen dieser Ar
beit wurde zur Quantifizierung und zum einfachen Vergleich der Halbzeugqualität ein
Qualitätsanalyseverfahren entwickelt und damit ein geeigneter FTS für weitere Pro
zessuntersuchungen ausgewählt. Zudem wurde eine FIFDM-Prototypenanlage ent
wickelt und aufgebaut. Mithilfe der thermischen Simulation des Extrusions- und Ab
kühlprozesses konnten thermische Prozessgrenzen auch für die 3D-Ablage im Raum
definiert werden. In einer umfassenden experimentellen Prozessanalyse wurde zu
dem untersucht, welche Prozessparameter einen Einfluss auf verschiedene Zielgrö
ßen der Prozessstabilität und Bauteilqualität besitzen. Ausgehend von den Erkennt
nissen aus dieser Arbeit wurden eine erste Einschätzung des Prozesspotentials vor
genommen und Vorschläge zur Prozessoptimierung formuliert.
3D printing enables automated and flexible production of complex 3D geometries
directly from a CAD model without the need for a component-specific tool. However,
the disadvantage, especially in the Additive Manufacturing (AM) of polymers, is the
low mechanical properties, which can be attributed to process-related challenges and
to a limited selection of processable materials. One way of improving the mechanical
properties of polymers is to combine them with reinforcing fibers. The highest rein
forcing effect for Fiber Reinforced Polymer Composites (FRPC) is achieved when the
fibers are continuously present in load direction. In order to develop their full poten
tial, FRPC must therefore be adapted as well as possible to the respective applica
tion. This complicates automated and efficient production, especially of more com
plex structures. The aim of the work was therefore to develop an AM process for con
tinuously fiber-reinforced polymers. This should increase the range of applications for
polymer-based AM processes and at the same time enable efficient and flexible pro
duction of complex FRPC structures. The developed process concept is based on 3D
printing extrusion processes for thermoplastics. In the so-called Fiber Integrated
Fused Deposition Modeling Process (FIFDM) already impregnated semi-finished
products are processed in the form of continuously fiber-reinforced thermoplastic
strands (FTS). In order to be able to freely adjust the fiber orientation, the strands can
be positioned in all spatial directions, not just layer by layer as is the case with con
ventional AM systems. This is realized by controlling the FTS temperature after ex
trusion. As part of this work, a quality analysis method was developed for quantifying
and comparing the semi-finished product quality and a suitable FTS was thus select
ed for further process investigations. In addition, a FIFDM prototype unit was devel
oped and set up. With the help of thermal simulation of the extrusion and cooling
process, thermal process limits could also be defined for the 3D placement in all spa
tial directions. In a comprehensive experimental process analysis, it was investigated
which process parameters have an influence on different target parameters of pro
cess stability and component quality. Based on the results of this work, an initial as
sessment of the process potential was made and proposals for process optimization
were formulated.
To achieve the Paris climate protection goals there is an urgent need for action in the energy sector. Innovative concepts in the fields of short-term flexibility, long-term energy storage and energy conversion are required to defossilize all sectors by 2040. Water management is already involved in this field with biogas production and power generation and partly with using flexibility options. However, further steps are possible. Additionally, from a water management perspective, the elimination of organic micropollutants (OMP) is increasingly important. In this feasibility study a concept is presented, reacting to energy surplus and deficits from the energy grid and thus providing the needed long-term storage in combination with the elimination of OMP in municipal wastewater treatment plants (WWTPs). The concept is based on the operation of an electrolyzer, driven by local power production on the plant (photovoltaic (PV), combined heat and power plant (CHP)-units) as well as renewable energy from the grid (to offer system service: automatic frequency restoration reserve (aFRR)), to produce hydrogen and oxygen. Hydrogen is fed into the local gas grid and oxygen used for micropollutant removal via upgrading it to ozone. The feasibility of such a concept was examined for the WWTP in Mainz (Germany). It has been shown that despite partially unfavorable boundary conditions concerning renewable surplus energy in the grid, implementing electrolysis operated with regenerative energy in combination with micropollutant removal using ozonation and activated carbon filter is a reasonable and sustainable option for both, the climate and water protection
Financing measures and incentive schemes for (existing and new) building owners can promote the sustainable settlement development of rural regions or municipalities and, in a wider sense, entire countries or cross-border regions. In order to be used on a broad scale, the concept of revolving funds must continue to be further developed. In this research, the concept of an advanced revolving housing fund (ARF) for building owners to support the sustainable development of rural regions and potential mechanisms are introduced. The ARF is designed to reflect impacts and challenges with regard to rural regions in Germany, Europe and beyond. Based on New Institutional Economics, the Theory of Spatial Organisms, an expert workshop, interviews and discussions and further literature research, the fundamentals for incentive schemes and the essential mechanisms and design aspects of the ARF are derived. This includes the principal structure and governance of a holding fund and several regional funds. Based on this, input parameters for the financial modelling of an ARF are presented as well as guiding elements for empirical testing to promote more research in this area. It is found that the ARF should have a regional focus and must be a comprehensive instrument of settlement development with additional informal and formal measures. The developed concept promises new impulses, in particular, for rural regions. It is proposed to test the concept by means of case studies in pioneer regions of different countries
In recent years, the Internet has become a major source of visual information exchange. Popular social platforms have reported an average of 80 million photo uploads a day. These images, are often accompanied with a user provided text one-liner, called an image caption. Deep Learning techniques have made significant advances towards automatic generation of factual image captions. However, captions generated by humans are much more than mere factual image descriptions. This work takes a step towards enhancing a machine's ability to generate image captions with human-like properties. We name this field as Affective Image Captioning, to differentiate it from the other areas of research focused on generating factual descriptions.
To deepen our understanding of human generated captions, we first perform a large-scale Crowd-Sourcing study on a subset of Yahoo Flickr Creative Commons 100 Million Dataset (YFCC100M). Three thousand random image-caption pairs were evaluated by native English speakers w.r.t different dimensions like focus, intent, emotion, meaning, and visibility. Our findings indicate three important underlying properties of human captions: subjectivity, sentiment, and variability. Based on these results, we develop Deep Learning models to address each of these dimensions.
To address the subjectivity dimension, we propose the Focus-Aspect-Value (FAV) model (along with a new task of aspect-detection) to structure the process of capturing subjectivity. We also introduce a novel dataset, aspects-DB, following this way of modeling. To implement the model, we propose a novel architecture called Tensor Fusion. Our experiments show that Tensor Fusion outperforms the state-of-the-art cross residual networks (XResNet) in aspect-detection.
Towards the sentiment dimension, we propose two models:Concept & Syntax Transition Network (CAST) and Show & Tell with Emotions (STEM). The CAST model uses a graphical structure to generate sentiment. The STEM model uses a neural network to inject adjectives into a neutral caption. Achieving a high score of 93% with human evaluation, these models were selected as the top-3 at the ACMMM Grand Challenge 2016.
To address the last dimension, variability, we take a generative approach called Generative Adversarial Networks (GAN) along with multimodal fusion. Our modified GAN, with two discriminators, is trained using Reinforcement Learning. We also show that it is possible to control the properties of the generated caption-variations with an external signal. Using sentiment as the external signal, we show that we can easily outperform state-of-the-art sentiment caption models.
In a (linear) parametric optimization problem, the objective value of each feasible solution is an affine function of a real-valued parameter and one is interested in computing a solution for each possible value of the parameter. For many important parametric optimization problems including the parametric versions of the shortest path problem, the assignment problem, and the minimum cost flow problem, however, the piecewise linear function mapping the parameter to the optimal objective value of the corresponding non-parametric instance (the optimal value function) can have super-polynomially many breakpoints (points of slope change). This implies that any optimal algorithm for such a problem must output a super-polynomial number of solutions. We provide a method for lifting approximation algorithms for non-parametric optimization problems to their parametric counterparts that is applicable to a general class of parametric optimization problems. The approximation guarantee achieved by this method for a parametric problem is arbitrarily close to the approximation guarantee of the algorithm for the corresponding non-parametric problem. It outputs polynomially many solutions and has polynomial running time if the non-parametric algorithm has polynomial running time. In the case that the non-parametric problem can be solved exactly in polynomial time or that an FPTAS is available, the method yields an FPTAS. In particular, under mild assumptions, we obtain the first parametric FPTAS for each of the specific problems mentioned above and a (3/2 + ε) -approximation algorithm for the parametric metric traveling salesman problem. Moreover, we describe a post-processing procedure that, if the non-parametric problem can be solved exactly in polynomial time, further decreases the number of returned solutions such that the method outputs at most twice as many solutions as needed at minimum for achieving the desired approximation guarantee.
This article is dedicated to the weight set decomposition of a multiobjective (mixed-)integer linear problem with three objectives. We propose an algorithm that returns a decomposition of the parameter set of the weighted sum scalarization by solving biobjective subproblems via Dichotomic Search which corresponds to a line exploration in the weight set. Additionally, we present theoretical results regarding the boundary of the weight set components that direct the line exploration. The resulting algorithm runs in output polynomial time, i.e. its running time is polynomial in the encoding length of both the input and output. Also, the proposed approach can be used for each weight set component individually and is able to give intermediate results, which can be seen as an “approximation” of the weight set component. We compare the running time of our method with the one of an existing algorithm and conduct a computational study that shows the competitiveness of our algorithm. Further, we give a state-of-the-art survey of algorithms in the literature.
Die Branche der Abwasser-, Abfall- und Entsorgungswirtschaft steht vor diversen neuen Herausforderungen, die insbesondere im Zuge der Digitalisierung und strengerer Anforderungen an die Recyclingquoten zu erwarten sind. Diese Erkenntnisse gehen aus der Analyse diverser Branchenreporte und Expertengespräche hervor. Um diese Entwicklungen näher zu beleuchten und daraus einen potenziellen Weiterbildungsbedarf zu erschließen, hat das Teilprojektteam der Hochschule Kaiserslautern zusätzlich eine Online-Branchenbefragung durchgeführt. Diese fokussiert neben dem aktuellen und prospektiven Personalbedarf auch die Notwendigkeit der Weiterbildung in spezifischen Sektoren sowie (zukünftige) Herausforderungen. Der vorliegende Arbeits- und Forschungsbericht umfasst die methodische Vorgehensweise der qualitativen und quantitativen Analysen sowie die Darstellung der jeweiligen Ergebnisse. Um den akuten Bedarfen nachzukommen und Herausforderungen entgegenzuwirken, wird eine Handlungsempfehlung für ein wissenschaftliches Weiterbildungsangebot an der Hochschule Kaiserslautern formuliert.
Laser-based powder bed fusion (L-PBF) is a promising technology for the production of near net–shaped metallic components. The high surface roughness and the comparatively low-dimensional accuracy of such components, however, usually require a finishing by a subtractive process such as milling or grinding in order to meet the requirements of the application. Materials manufactured via L-PBF are characterized by a unique microstructure and anisotropic material properties. These specific properties could also affect the subtractive processes themselves. In this paper, the effect of L-PBF on the machinability of the aluminum alloy AlSi10Mg is explored when milling. The chips, the process forces, the surface morphology, the microhardness, and the burr formation are analyzed in dependence on the manufacturing parameter settings used for L-PBF and the direction of feed motion of the end mill relative to the build-up direction of the parts. The results are compared with a conventionally cast AlSi10Mg. The analysis shows that L-PBF influences the machinability. Differences between the reference and the L-PBF AlSi10Mg were observed in the chip form, the process forces, the surface morphology, and the burr formation. The initial manufacturing method of the part thus needs to be considered during the design of the finishing process to achieve suitable results.
The 22 wt.% Cr, fully ferritic stainless steel Crofer®22 H has higher thermomechanical
fatigue (TMF)- lifetime compared to advanced ferritic-martensitic P91, which is assumed to be caused
by different damage tolerance, leading to differences in crack propagation and failure mechanisms.
To analyze this, instrumented cyclic indentation tests (CITs) were used because the material’s
cyclic hardening potential—which strongly correlates with damage tolerance, can be determined
by analyzing the deformation behavior in CITs. In the presented work, CITs were performed for
both materials at specimens loaded for different numbers of TMF-cycles. These investigations show
higher damage tolerance for Crofer®22 H and demonstrate changes in damage tolerance during
TMF-loading for both materials, which correlates with the cyclic deformation behavior observed in
TMF-tests. Furthermore, the results obtained at Crofer®22 H indicate an increase of damage tolerance
in the second half of TMF-lifetime, which cannot be observed for P91. Moreover, CITs were performed
at Crofer®22 H in the vicinity of a fatigue crack, enabling to locally analyze the damage tolerance.
These CITs show differences between crack edges and the crack tip. Conclusively, the presented
results demonstrate that CITs can be utilized to analyze TMF-induced changes in damage tolerance.
Designing exotic structures in low dimensions is key in today’s quest to tailor novel quantum states
in materials with unique symmetries. Particularly intriguing materials in this regard are low
dimensional aperiodic structures with non-conventional symmetries that are otherwise forbidden
in translation symmetric crystals. In our work, we focus on the link between the structural and
electronic properties of aperiodically ordered aromatic molecules on a quasicrystalline surface,
which has largely been neglected so far. As an exemplary case, we investigate the self-assembly and
the interfacial electronic properties of the nano-graphene-like molecule coronene on the bulk
truncated icosahedral (i) Al–Pd–Mn quasicrystalline surface using multiple surface sensitive
techniques. We find an aperiodically ordered coronene monolayer (ML) film on the i-Al–Pd–Mn
surface that is characterized by the same local motifs of the P1 Penrose tiling model as the bare
i-Al–Pd–Mn surface. The electronic valence band structure of the coronene/i-Al–Pd–Mn system
is characterized by the pseudogap of the bare i-Al–Pd–Mn, which persists the adsorption of
coronene confirming the quasiperiodic nature of the interface. In addition, we find a newly formed
interface state of partial molecular character that suggests an at least partial chemical interaction
between the molecule and the quasicrystalline surface. We propose that this partial chemical
molecule–surface interaction is responsible for imprinting the quasicrystalline order of the surface
onto the molecular film.
Purpose
We investigated the cytosolic and membrane-associated contents of polyphenols after 4 hours of incubation (50 μM of each polyphenol) in the colon carcinoma cell line T84 using a novel, rapid, and convenient method based on permeabilization of the cell membrane using digitonin. The colon carcinoma cell line was used to investigate the intestinal uptake of polyphenols present in apple products.
Recent Findings
The results showed that hydroxycinnamic acids (caffeic and 5-caffeoylquinic acid) were only detected in the cytosolic fractions. In contrast, 0.3 to 8.2% of the initial concentrations (50 μM) of the flavonoids phloretin, quercetin, phloretin 2′-O-glucoside, and quercetin 3-O-rhamnoside were found in the membrane-associated fractions. In the cytosolic fractions, 0.2–2.9% of these compounds were detected, corresponding to 25 to 40% of the total cell-associated (cytosolic plus membrane-associated fractions) polyphenol content.
Summary
Our results showed that after uptake, polyphenols were present in the cytosolic fraction of the cells as well as associated with the cell membrane. The presented method provides a useful in vitro tool for determining biologically active compounds in cellular fractions.
Die Monomere Ethen und Propen gehören zu den wichtigsten Grundbausteinen der chemischen Industrie. Die zahlreichen Einsatzmöglichkeiten dieser kurzkettigen Alkene sorgen dafür, dass der Bedarf stetig steigt. Um diesen decken zu können, werden Olefine durch diverse Syntheserouten großtechnisch hergestellt. Das Steamcracken und katalytische Cracken (FCC) von Naphtha oder Ethan gehören zu den am meisten verbreiteten Verfahren, um leichte Olefine zu synthetisieren. Diese Technologien sind allerdings mit einigen Nachteilen, wie zum Beispiel einem relativ hohen Energieverbrauch und einer geringen Selektivität für bestimmte Olefine verbunden. Endliche Erdölreserven und steigende Erdölpreise führen schlussendlich zur Entwicklung neuartiger Synthesemethoden für leichte Olefine über nachhaltige Rohstoffe speziell für Produktionsanlagen, die nicht völlig rückintegriert zu Raffinerien oder Steamcracker sind.
Ethen, Propen und Butene können auch selektiv aus den korrespondierenden Alkanen durch Dehydrierung erhalten werden. Der Vorteil der selektiven Dehydrierung besteht darin, dass kein kompliziertes Produktgemisch vorliegt und die Reaktion somit nicht mit einer aufwendigen Produkttrennung verbunden ist. Die Dehydrierung von Propan zu Propen ist mit einer erheblichen Wertsteigerung am Markt verbunden und wird bereits vielfältig eingesetzt. Ethan kommt als große Nebenkomponente im preiswerten Erdgas vor. Es ist daher interessant, Ethan zu dehydrieren und anschließend in situ zu Propen und Butenen zu dimerisieren. Neben der Dehydrierung von Ethan kann Ethen aber auch aus Ethanol hergestellt werden.
Durch die Dehydratisierung von Ethanol kann Ethen aus nachwachsenden Rohstoffen, wie zum Beispiel aus zucker- oder stärkehaltigen Materialien oder auch aus Lignocellulose hergestellt werden. Über Fermentation und anschließende Aufreinigung ist Bioethanol leicht aus Korn, Zuckerrohr und Cellulose zugänglich.
Zeolithe werden als azide, feste Katalysatoren in der chemischen Industrie eingesetzt. Diverse Porenarchitekturen und unterschiedliche Dimensionalitäten ermöglichen eine sehr gute Steuerung der Selektivität in Bezug auf ein erwünschtes Produkt. Über das Silizium zu Aluminium-Verhältnis kann die Anzahl der sauren Zentren angepasst und können säurekatalysierte Reaktionen beeinflusst werden. Die einfache Herstellung sowie Abtrennung des Katalysators aus dem Produktgemisch sorgen dafür, dass feste Katalysatoren, insbesondere Zeolithe, einen hohen Stellenwert in der Industrie haben. Deshalb werden sie in dieser Arbeit als Katalysatoren verwendet.
Der Schwerpunkt dieser Arbeit liegt in der Untersuchung von Verfahren zur Produktion leichter Olefine aus Ethan und Ethanol. Je nachdem welche Reaktionsbedingungen bei der oder bei der Dehydrierung gewählt werden, ist Ethen das Hauptprodukt. Die anschließende Oligomerisierung von Ethen zu Propen, Butenen oder sogar zu BTX-Aromaten ist hingegen eine Herausforderung. Im ersten Teil dieser Arbeit wird Dehydratisierung die Dehydratisierung von Ethanol zu leichten Olefinen untersucht. Hierbei wird die Abhängigkeit des Produktspektrums von Zeolithen mit unterschiedlicher Dimensionalität und Porengröße ermittelt. Anschließend wird die Abhängigkeit der Olefinausbeute von der Partikelgröße untersucht und das Produktspektrum der Dehydrierung von Ethan über den Einsatz ausgewählter Metalle optimiert.
Die eingesetzten Zeolithe werden je nach gewünschter Porenarchitektur und Dimensionalität synthetisiert und entsprechend physikochemisch charakterisiert.
Der Ausbruch der Covid-19-Pandemie hatte massive Auswirkungen auf die Weiterbildungsbrache und die Folgen sind bisher noch nicht abzuschätzen. Durch die Absage/Aussetzung aller Präsenzveranstaltungen sowie Kontaktbeschränkungen für Mitarbeitende der Weiterbildungseinrichtungen zur Eindämmung des Virus im März diesen Jahres, mussten ad-hoc neue Grundlagen für die Arbeit geschaffen werden. Im Rahmen der vorliegenden Befragungsergebnisse von 10 Volkshochschulen in Rheinland-Pfalz im Mai/Juni wird beispielhaft nachgezeichnet, wie die Einrichtungen mit den Einschränkungen der Pandemie umgegangen sind. Dabei wird insbesondere auf die Digitalisierung im Bereich der Angebote und der Zusammenarbeit sowie die Folgen der aktuellen Krise eingegangen. Die Ergebnisse zeigen dabei, dass die Einschränkungen sowohl als existenzielle Bedrohung, als auch Chance für eine Weiterentwicklung der Einrichtungen gesehen werden. Dabei hat die Digitalisierung einen deutlichen Schub erfahren. Eine grundlegende Veränderung der Ziele und Strategien der Einrichtungen ist jedoch nicht zu erkennen.
Die Masterarbeit unternimmt den Versuch aufzuzeigen, wie der Street Artist Banksy Reflexionsprozesse im und über den Kunstbetrieb auslöst. Dies wird im Sinne der Systemtheorie Luhmanns verstanden, bei der Reflexion als Selbstreferenz sozialer Systeme gilt und dann zutage tritt, wenn das System eine Irritation erfährt.
Banksys Irritationen entstehen durch unterschiedliche Aktionen und Verkaufssettings, beispielsweise durch das eigenmächtige Aufhängen seiner Bilder in einigen der renommiertesten Museen der Welt oder durch das Schreddern des Bildes Girl with Balloon während einer Auktion bei Sotheby’s. Dabei wird betrachtet, wie Banksy die häufig irrationalen Preismechanismen des Kunstmarktes motivisch im Bild und durch seine Beiträge thematisiert, wie z.B. mit einem Verkaufsstand im New Yorker Central Park.
In der Erörterung dieses aktuellen Themas wird deutlich, wie er sich zunehmend vom reinen Graffiti Sprayer/Street Artist zum grenzüberschreitenden Aktionskünstler entwickelt. Dabei nimmt er selbst verschiedene genreübergreifende Rollen, wie z.B. die des Museumskünstlers, Filmregisseurs, Art Handlers, Gutachters, Kurators, Auktionators oder Kunstvermittlers im Kunstsystem ein, die sonst von mehreren vermittelnden Personen besetzt werden.
Es wird sowohl ersichtlich, wie Banksy selbst ein dynamisches Künstlerbild mit den Mitteln der Street Art entwickelt, als auch, wie die Maskierung und die Verwendung eines Pseudonyms, auch für die Beziehung zu den Betrachter*innen, von großer Bedeutung ist und wie die Absenz von Informationen für eine Sogwirkung in der Rezeption sorgt.
Mit der in der Forschung noch wenig beachteten Fragestellung “Wie setzt Banksy Reflexionsprozesse über den Kunstbetrieb in Gang” wird in der Masterarbeit aufgezeigt, wie Reflexionsprozesse durch Irritationen entstehen, die Banksy durch seine originellen künstlerischen Interventionen erzeugt. Aus dem Vergleich seiner künstlerischen Handlungen auf und abseits der Straße, wie z.B. im institutionellen Rahmen von Museen, wird dies deutlich.
Vor dem Hintergrund, dass kleine und mittlere Flughäfen strukturell defizitär und zumindest in Bezug auf die Infrastrukturfinanzierung auf öffentliche Fördermittel angewiesen sind , stellen sich unweigerlich die Fragen, inwieweit die jeweiligen staatlichen Unterstützungsmaßnahmen mit den beihilferechtlichen Regelungen kompatibel sind bzw. mit welchen Konsequenzen ein Regionalflughafen im Falle eines erwiesenen Beihilfeverstoßes rechnen muss. Die vorliegende Arbeit hat sich deshalb zum Ziel gesetzt, die Bedeutung des EU-Beihilferechts für Regionalflughäfen in Deutschland aufzuzeigen.
Hierfür ist zunächst zu klären, wann ein Flughafen als Regionalflughafen klassifiziert wird, welche Besonderheiten Regionalflughäfen in Deutschland grundsätzlich aufweisen und mit welchen Herausforderungen sie in der heutigen Zeit konfrontiert sind. Exemplarisch soll sich dabei im Besonderen dem Regionalflughafen Frankfurt-Hahn (Rheinland-Pfalz) angenommen werden, da dieser bereits mehrfach das Zielobjekt beihilferechtlicher Untersuchungen sowie diverser Gerichtsverfahren war.
Im weiteren Verlauf ist der Frage nachzugehen, unter welchen Gegebenheiten der Beihilfetatbestand bei Regionalflughäfen (insb. am Beispiel Frankfurt-Hahn) erfüllt ist und inwiefern solche Beihilfen akzeptiert bzw. für mit dem Binnenmarkt vereinbar erklärt werden können. In diesem Zuge soll auch ein Überblick über die einschlägigen Normen (Primär-/ Sekundärrecht, „soft law“) sowie die relevante Entscheidungspraxis der Kommission bzw. der Unionsgerichte gegeben werden.
Schließlich ist zu untersuchen, welchen Risiken der Regionalflughafen Frankfurt-Hahn momentan aufgrund der beihilferechtlichen Regelungen ausgesetzt ist und welche Folgen mit rechtswidrig gewährten Beihilfen verbunden sein können. Abschließend soll sodann skizziert werden, worauf ein Regionalflughafen in Deutschland zu achten hat, um sich in das Korsett, welches das EU-Beihilferecht diesem Sektor verpasst hat, zu zwängen.
Nach einer Zusammenfassung der gewonnenen Erkenntnis ist zuletzt beabsichtigt, auf offene, noch zu erörternde Fragestellungen hinzuweisen.
Diese Arbeit verbessert einige Prozessschritte zur Auslegung hochbeanspruchter Bauteile aus
endlosfaserverstärkten Faser-Thermoplast-Verbunden am Beispiel von CF-PA6 Mehrschichtverbunden.
Hierzu werden neue Verfahren zur Konditionierung hygroskopischer Werkstoffe und der
Bestimmung des relativen Faservolumenanteils entwickelt, wodurch der Aufwand zur Werkstoffcharakterisierung
signifikant reduziert werden kann.
Durch Versuche an CF-PA6 Schichten wird das mechanische Verhalten des Werkstoffs hinsichtlich
dem Spannungs-Verzerrungs- und Bruchverhalten analysiert. Dazu werden speziell die Unterschiede
zu Faser-Duroplast-Verbunden durch Berechnungen auf mikromechanischer Ebene herausgearbeitet.
Zusätzlich wird der Einfluss des verwendeten Verarbeitungsverfahrens (Tapelegen) auf
die mechanischen Werkstoffkennwerte charakterisiert.
Auf Basis der gewonnenen Erkenntnisse wird eine neue Werkstoffroutine entwickelt und in ein Berechnungsprogramm
zur Analyse mechanischer Strukturen mit Hilfe der Finiten Elemente Methode
implementiert. Dadurch kann unter anderem das nichtlineare Verhalten von Faser-Thermoplast-
Verbunden unter großen Verzerrungen und die Degradation einzelner Schichten eines Mehrschichtverbunds
abgebildet werden. Ein Vergleich der Ergebnisse von Versuchen an Mehrschichtverbunden
und der Berechnung zeigt gute Übereinstimmung und die Leistungsfähigkeit der Routine auf Bauteilebene.
This work improves the process steps for the design of highly stressed components made of continuous
fiber-reinforced thermoplastics using as example CF-PA6 multilayer composites. For this
purpose, new methods for conditioning hygroscopic materials and determining the relative fiber
volume fraction were developed. This reduces the expense of material characterization significantly.
Tests on CF-PA6 layers are used to analyze the mechanical behavior of the material in terms of
stress-strain and fracture behavior. In particular, the differences to fiber-reinforced thermosets are
highlighted by using calculations on a micromechanical level. Additionally, the influence of the
processing method (tape laying) on the mechanical material properties is characterized.
Based on the knowledge gained, a new material routine is developed and implemented into a
calculation program for the analysis of mechanical structures using the finite element method.
Through this work it is now possible to calculate nonlinear behavior of fiber-reinforced thermoplastics
under large distortions and the degradation of individual layers of a multi-layer composite.
A comparison of experimental data with the calculation of multilayer composites shows good
agreement and the performance for the application on a component-level.
Maschinenbetten und -gestelle werden vorwiegend aus Grauguss und als Stahlschweißkonstruktion gefertigt. Aufgrund technischer und wirtschaftlicher Vorteile haben sich seit den 80er Jahren zusätzlich Mineralguss oder epoxidharzgebundener Polymerbeton etabliert. Seit mehreren Jahren wird Ultra High Performance Concrete – UHPC – erfolgreich bei Werkzeugmaschinen eingesetzt. Auf diesen Grundgestellen aus Beton werden neben Gehäusen, Getrieben, Werkzeughaltern, Trafos usw. vor allem die beweglichen Teile der Maschine befestigt. Gängige Praxis hierfür ist die Verwendung von Linearführungsschienen.
Diese Arbeit untersucht die vorgespannte, geschraubte Befestigung der Linearführung auf geschliffenen Betonoberfläche mit Hilfe von speziellen Gewindehülsen. In rechnerischen Voruntersuchungen mittels der FE-Methode wurde der Kraftfluss von der Linearführung über die Befestigungsschraube zur Gewindehülse bis in den Betonuntergrund untersucht und darauf aufbauend ein Versuchsprogramm festgelegt. Probekörper aus hochfestem Beton des Marktführers wurden hergestellt und experimentell untersucht. Nach der Auswertung der Versuche wurde ein Bemessungskonzept entwickelt und in Form einer schrittweisen Anleitung festgehalten. Mehrere Beispiele zeigen die Anwendung des Verfahrens.
Berechnungen, Versuchskonzeption, Durchführung der Versuche, Versuchsauswertung sowie die Entwicklung des Bemessungskonzeptes erfolgten durch den Verfasser. Die Versuche wurden in seiner Anwesenheit in seinem Betrieb, in spezialisierten Unternehmen mit entsprechender Geräteausstattung und zum größten Teil an der TU Kaiserslautern durchgeführt. Die hier betrachteten Untersuchungen und Bemessungsvorschläge betrachten ausschließlich Maschinenbauteile und dürfen nicht auf Situationen im Bauwesen übertragen werden.
Ein ressourcenschonender Umgang mit Rohstoffen ist eines der vorrangigen Ziele des 21. Jahrhunderts und erfährt immer mehr Aufmerksamkeit in der Öffentlichkeit. Einen nicht unerheblichen Beitrag zur Reduzierung des Ressourcenbedarfs kann der Einsatz von Hohlkörpern in Stahlbetondecken beisteuern. Durch den verminderten Betoneinsatz wird weniger CO2 bei der für den Beton erforderlichen Zementherstellung emittiert. Einen weiteren positiven Aspekt für den Einbau von Hohlkörpern in Stahlbetondecken stellt das deutlich reduzierte Eigengewicht dar, welches größere Spannweiten bei gleicher Deckenstärke ermöglicht.
Während sich die Biegetragfähigkeit einer Stahlbetonhohlkörperdecke lediglich geringfügig von der Biegetragfähigkeit einer massiven Stahlbetondecke unterscheidet, wird die Querkrafttragfähigkeit durch den Einbau der Hohlkörper deutlich reduziert. Die Querkrafttragfähigkeit der Hohlkörperdecke liegt bei den bislang im Bauwesen eingesetzten Hohlkörpern bei ungefähr 50 % der Querkrafttragfähigkeit einer baugleichen Vollmassivdecke, so dass die Hohlkörper in den Bereichen einer Decke, in denen hohe Querkräfte vorliegen, nicht eingebaut werden dürfen. Dies führt zu einer deutlichen Einschränkung des Einsatzbereiches der Hohlkörper. Da die Hohlkörperformen bislang keinem Optimierungsprozess hinsichtlich der Querkrafttragfähigkeit unterzogen wurden, sind die in der Baupraxis verwendeten Hohlkörperformen keinesfalls als optimal anzusehen.
In dieser Arbeit wird mit Hilfe eines mehrkriteriellen Optimierungsprozesses, basierend auf der Methode der Genetischen Programmierung, eine Hohlkörperform identifiziert, die hinsichtlich der beiden konträren Optimierungskriterien „hohe Querkrafttragfähigkeit“ und „hohe Betonvolumenverdrängung“ einen guten Kompromiss darstellt. Neben den numerischen FEM-Berechnungen zur Ermittlung der Querkrafttragfähigkeit, welche u.a. auch während des Optimierungsprozesses zur Anwendung kommen, wird die Querkrafttragfähigkeit von Plattenstreifen mit den optimierten Hohlkörpern zusätzlich in Bauteilversuchen überprüft. Um ein ganzheitlich durchdachtes neues Hohlkörperdeckensystem zu entwickeln, werden neben der Optimierung der Querkrafttragfähigkeit auch zahlreiche weitere Untersuchungen zur Optimierung des Materialeinsatzes sowie zur Transport- und Einbausituation durchgeführt.
Des Weiteren bildet die Entwicklung eines mechanisch begründeten Bemessungsmodells zur Ermittlung der Querkrafttragfähigkeit einer Hohlkörperdecke einen wesentlichen Bestandteil der vorliegenden Arbeit. Um ein abgesichertes Modell zu erstellen, welches die Querkrafttragfähigkeit realitätsnah abbildet, sind fundierte Kenntnisse zum Querkrafttragverhalten erforderlich. Dafür werden neben einer mittels der Finite Elemente Methode durchgeführten umfangreichen Parameterstudie, in der zahlreiche Einflussfaktoren auf die Querkrafttragfähigkeit von Hohlkörperdecken untersucht werden, ergänzend Bauteilversuche mit variierender Hohlkörperhöhenlage durchgeführt.
Acrylamid ist eine Lebensmittelkontaminante, welche durch die sogenannte Maillard-Reaktion im Lebensmittel gebildet wird. Acrylamid entsteht insbesondere bei der Zubereitung kohlenhydratreicher hocherhitzer Lebensmittel, wie z.B. Pommes frites, Chips, Cerealien und ähnlichen Produkten. Die mittlere tägliche Exposition, durch den Verzehr von Lebensmittel liegt bei ca. 0,4–1,9 µg/kg Körpergewicht und Tag. Darüber hinaus entsteht Acrylamid in Folge von Pyrolyseprozessen, weshalb Tabakrauchen einen weiteren Expositionsweg darstellt. Die Menge des dabei entstehenden Acrylamids im Hauptstrom von Zigarettenrauch liegt bei 1,1–2,3 µg/Zigarette. In Studien erwies sich die Substanz als reproduktions- und gentoxisch, sowie kanzerogen beim Nager und zudem als neurotoxisch sowohl im Nager als auch beim Menschen. Bereits 1994 wurde Acrylamid von der International Agency for Research on Cancer (IARC) als wahrscheinlich krebserzeugend für den Menschen (Gruppe 2A) eingestuft. Im Zuge des Fremdstoffmetabolismus wird Acrylamid in der Leber zu Glycidamid umgesetzt. Katalysiert wird die Reaktion durch das Enzym CYP2E1 ist. Als reaktives Epoxid kann Glycidamid an die DNA binden, wodurch das in vorliegender Arbeit behandelte N7-(2-Carbamoyl-2-Hydroxyethyl)-guanin (N7-GA-Gua), gebildet wird. Dieses DNA-Addukt bildet das Hauptaddukt der Reaktion aus GA und DNA-Bausteinen.
Ziel der vorliegenden Arbeit war es, eine hochsensitive UHPLC-ESIpos-MS/MS-Methode zu entwickeln, um damit anschließend eine mögliche Hintergrundbelastung mit Acrylamid anhand des N7-GA-Gua-Addukts sowohl im Tier als auch im Menschen nachzuweisen und damit eine bessere Datenlage über die Substanz zu erhalten.
Zu Beginn dieser Arbeit wurden Synthesen der für die chromatographische Analytik benötigten Verbindungen durchgeführt. Hierbei konnten erstmals Glycidamid Kristalle erhalten und untersucht werden. In nachfolgenden in-vitro-Experimenten wurden die Zytotoxizität von Acrylamid und dessen Metaboliten mittels Resazurin-Reduktionstest, sowie die Bildung von N7-GA-Gua in primären Rattenhepatozyten (pRH) nach Inkubation mit Acrylamid, untersucht. Im Resazurin Test erwies sich Acrylamid in Konzentrationen bis zu 2500 µM als nicht zytotoxisch und konnte daher zur Inkubation der Zellen mit Konzentrationen bis zu 2000 µM Acrylamid eingesetzt werden. Im anschließend durchgeführten Inkubationsversuch, bei dem die Zellen mit 2–2000 µM Acrylamid über 1, 16 oder 24 h inkubiert wurden, konnten in allen Testkonzentrationen N7-GA-Gua-Addukte bestimmt werden. Dank der im Rahmen dieser Arbeit entwickelten sensitiven analytischen Methode, konnten N7-GA-Gua-Addukte auch in den Organen (Lunge, Niere und teilweise Leber) vom Nager (C57BL/6jRj- und BKS(D) Leprdb/JOrLR Maus und Wistar-Ratte) bestimmt werden. Geschlechts- oder organabhängige Effekte konnten nicht festgestellt werden. Einzig die Anzahl an N7-GA-Gua lag in den C57BL/6jRj-Mäusen signifikant über denen der Wistar-Ratten. In der durchgeführten Humanstudie konnten im Blut von 48 der 60 Probanden Addukte nachgewiesen werden. Signifikante Einflüsse der Lebensführung, des Alters und der Ernährung der Probanden auf den Hintergrund-Adduktgehalte wurden nicht festgestellt; es ergab sich lediglich eine schwache Korrelation zwischen der Anzahl an N7-GA-Gua und dem Gewicht bzw. dem BMI der Studienteilnehmer.
Insgesamt liefert die vorliegende Arbeit wichtige Erkenntnisse über die Gehalte an N7-GA-Gua in vitro in primären Rattenhepatozyten, als auch in vivo in Organen von Ratte und Maus, sowie im Blut des Menschen. Die Ergebnisse tragen zur Verbesserung der Datenlage über Acrylamid bei. Insbesondere die Ergebnisse der durchgeführten Humanstudie liefern Einblicke in die Hintergrundbelastung an N7 GA-Gua im Querschnitt der Bevölkerung und können daher auch für die Risikobetrachtung von Acrylamid sowie zur Ableitung weiterer Forschungsarbeiten und Studien von Interesse sein.
Natürliche Inhaltstoffe von Lebensmitteln können ein gentoxisches Potential haben. Ein Beispiel
dafür sind Phenylpropanoiden, welche als sekundäre Pflanzeninhaltsstoffe in einer Vielzahl von
Gewürz- und Kräuterpflanzen vorkommen. Zu den prominenten Vertretern dieser Stoffe gehören
Estragol und Methyleugenol. Beide allylische Verbindungen wirken hepatokanzerogen im
Tierversuch, wobei als ultimales Kanzerogen die Bildung eines Carbeniumions infolge einer
spontanen Abspaltung eines Sulfatrestes nach Hydroxylierung und anschließender Sulfonierung
der Seitenkette postuliert wird. Dieses Carbeniumion ist in der Lage mit Nukleophilen wie Proteinen
und DNA Addukte zu bilden. α‐ und β‐Asaron nehmen im Gesamtkontext der Phenylpropene eine
Sonderstellung ein, denn im Gegensatz zu anderen 1‑propenylischen Phenylpropenen wie Anethol,
Methylisoeugenol und Isosafrol sind sie wie die allylischen Verbindungen Estragol und
Methyleugenol kanzerogen im Tierversuch.
Zu Beginn der vorliegenden Promotionsarbeit war der Metabolismus von α‐ und β‑Asaron
weitestgehend unbekannt. Aus früheren Studien leitete sich die Hypothese ab, dass die beiden
1‑propenylischen Asarone, nicht wie die allylischen Vertreter über Hydroxylierung und
anschließender Sulfonierung metabolisiert und somit aktiviert werden, sondern eine Epoxidierung
der Seitenkette zu reaktiven Metaboliten führen könnte. Nachdem das Metabolitenspektrum von
α‐ und β‐Asaron aufgeklärt wurde und die Epoxide als verantwortlich für das mutagene Potential
der Verbindungen im Ames-Fluktuationstest identifiziert werden konnten, sollte die Frage der
DNA-Adduktbildung in primären Rattenhepatozyten beantwortet werden. Hierfür wurde die
Reaktivität der Asaronepoxide gegenüber 2′-Desoxynukleosiden untersucht und die gebildeten
Addukte charakterisiert. In einer in-vitro-Studie konnte mit Hilfe einer sensitiven UHPLC-MS/MSMethode
ein konzentrationsabhängiger Zusammenhang zu den gebildeten DNA-Addukten
festgestellt werden. Weiter wurden in Untersuchungen zur Zeitabhängigkeit Hinweise auf Reparatur
der gebildeten Addukte gefunden.
Im Rahmen dieser Arbeit wurden 2,6-Bis(pyrazol-3-yl)pyridinliganden dargestellt und die korrespondierenden Rutheniumkomplexe hinsichtlich ihrer Aktivität in homogenkatalytischen Hydrierungs- und Dehydrierungsreaktionen untersucht. Durch Anwendung des Prinzips von Le Chatelier konnte die katalytische Transferhydrierung verschiedener Substrate mit Isopropanol, dem gängigen Lösungsmittel und Wasserstoffdonor dieser Umsetzung, optimiert werden. Zusätzlich gelang die Substitution dieses sekundären Alkohols gegen preiswertes Ethanol, welches aus nachwachsenden Rohstoffen gewonnen werden kann. Dabei nahm die rasche Entfernung des gebildeten Acetaldehyds aus dem Gleichgewicht eine Schlüsselrolle zur Vermeidung unerwünschter Nebenreaktionen und zum Erzielen hoher Produktausbeuten ein. Selektive Funktionalisierungen der Katalysatorstruktur lieferten grundlegende Einblicke in die Struktur-Eigenschafts-Beziehungen des Systems und trugen somit zu dessen Verständnis bei. Unter Berücksichtigung ökonomischer Aspekte wurden darüber hinaus zwei praktische, sehr effiziente Varianten der Ley-Griffith-Oxidation zur Dehydrierung unterschiedlicher Alkohole und Amine erarbeitet. Der zugrundeliegende Reaktionsmechanismus wurde anhand von DFT-Rechnungen eingehend erforscht.
Using molecular dynamics simulations, the adsorption and diffusion of doxorubicin drug molecules in boron nitride nanotubes are investigated. The interaction between doxorubicin and the nanotube is governed by van der Waals attraction. We find strong adsorption of doxorubicin to the wall for narrow nanotubes (radius of 9 Å). For larger radii (12 and 15 Å), the adsorption energy decreases, while the diffusion coefficient of doxorubicin increases. It does, however, not reach the values of pure water, as adsorption events still hinder the doxorubicin mobility. It is concluded that nanotubes wider than around 4 nm diameter can serve as efficient drug containers for targeted drug delivery of doxorubicin in cancer chemotherapy.
Here we describe a weakly interacting Bose gas on a curved smooth manifold, which is embedded in the three-dimensional Euclidean space. To this end we start by considering a harmonic trap in the normal direction of the manifold, which confines the three-dimensional Bose gas in the vicinity of its surface. Following the notion of dimensional reduction as outlined in [L Salasnich et al, Phys. Rev. A 65, 043614 (2002)], we assume a large enough trap frequency so that the normal degree of freedom of the condensate wave function can be approximately integrated out. In this way we obtain an effective condensate wave function on the quasi-two-dimensional surface of the curved manifold, where the thickness of the cloud is determined self-consistently. For the particular case when the manifold is a sphere, our equilibrium results show how the chemical potential and the thickness of the cloud increase with the interaction strength. Furthermore, we determine within a linear stability analysis the low-lying collective excitations together with their eigenfrequencies, which turn out to reveal an instability for attractive interactions.
Clean silica surfaces have a high surface energy. In consequence, colliding silica nanoparticles will stick rather than bounce over a wide range of collision velocities. Often, however, silica surfaces are passivated by adsorbates, in particular water, which considerably reduce the surface energy. We study the effect of surface hydroxylation on silica nanoparticle collisions by atomistic simulation, using the REAX potential that allows for bond breaking and formation. We find that the bouncing velocity is reduced by more than an order of magnitude compared to clean nanoparticle collisions
Reactive bubble columns are omnipresent in the chemical industry. The layout of these columns is still limited by correlations and therefore improved simulation techniques are required to describe the complex hydrodynamics/reaction interaction. In this work, we focus on the numerical and experimental study of the viscosity influence on bubble motion and reaction using an Euler-Lagrange framework with an added oscillation and reaction model to bring the column layout base closer to a predictive level. For comparison and validation, experimental data in various water-glycerol solutions was obtained in a cylindrical bubble column at low gas hold-up, where the main parameters such as bubble size, motion, and velocities were detected. Glycerol leads thereby to a change in viscosity and surface tension. Further, the surface tension was modified by addition of a surfactant. The bubble oscillating motion in low to higher viscosity could be described using an Euler-Lagrange framework and enables a description of industrial bubble flows. In addition, the simulations were in good agreement concerning reactive mass transfer investigations at higher viscosity of the liquid which led to an overall lower mass transfer compared to the cases with lower viscosity.
Strength training in youth soccer has both a preventive and a
sports-specific component. Whole-body electromyostimulation
(WB-EMS) could represent an interesting time-saving add-on to
classical strength exercises in performance-oriented soccer. The
objective of this study was to find out whether a 10-week superimposed
WB-EMS training might have a more positive impact on
strength parameters in male youth elite soccer players than regular
athletic strength exercises alone. A total of 30 male youth soccer
players from a youth academy aged 15 to 17 years participated
in the study. Before and after the intervention, the isometric extension
and flexion forces of trunk and knee, and the hip abduction
and adduction forces were tested. Twelve players (control
group) absolved a conventional 20-minute strength training once
a week for a period of ten weeks. Eighteen players absolved the
same exercises but with superimposed WB-EMS. Blood creatine
kinase concentration was measured for training control. ANOVAs,
Friedman tests and post hoc t-tests were calculated (p =
0.05) to examine the strength development during the training period
between the groups. While we could not find significant
strength increases in the leg, hip and trunk muscles in the control
group (<4%), the strength of the WB-EMS group improved significantly
in 4 of the 6 muscle groups tested. In this group, the
strength of knee flexors increased significantly by 20.68 ±
21.55%, knee extensors by 31.43 ± 37.02%, hip adductors by
21.70 ± 12.86% and trunk flexors by 33.72 ± 27.43%. The rates
of strength increase are partly in line with other studies, partly
clearly higher, which might be explained by the athletically active
target group. A 10-week superimposed WB-EMS training improves
the strength of certain leg, hip and trunk muscles in male
adolescent elite soccer players to a greater extent than a pure athletic
strength training of the same duration.
Red fruits and their juices are rich sources of polyphenols, especially anthocyanins.
Some studies have shown that such polyphenols can inhibit enzymes of the carbohydrate metabolism,
such as α-amylase and α-glucosidase, that indirectly regulate blood sugar levels. The presented
study examined the in vitro inhibitory activity against α-amylase and α-glucosidase of various
phenolic extracts prepared from direct juices, concentrates, and purees of nine different berries which
differ in their anthocyanin and copigment profile. Generally, the extracts with the highest phenolic
content—aronia (67.7 ± 3.2 g GAE/100 g; cyanidin 3-galactoside; chlorogenic acid), pomegranate
(65.7 ± 7.9 g GAE/100 g; cyanidin 3,5-diglucoside; punicalin), and red grape (59.6 ± 2.5 g GAE/100 g;
malvidin 3-glucoside; quercetin 3-glucuronide)—showed also one of the highest inhibitory activities
against α-amylase (326.9 ± 75.8 µg/mL; 789.7 ± 220.9 µg/mL; 646.1 ± 81.8 µg/mL) and α-glucosidase
(115.6 ± 32.5 µg/mL; 127.8 ± 20.1 µg/mL; 160.6 ± 68.4 µg/mL) and, partially, were even more potent
inhibitors than acarbose (441 ± 30 µg/mL; 1439 ± 85 µg/mL). Additionally, the investigation of single
anthocyanins and glycosylated flavonoids demonstrated a structure- and size-dependent inhibitory
activity. In the future in vivo studies are envisaged.
Although today’s bipeds are capable of demonstrating impressive locomotion skills, in many aspects, there’s still a big gap compared to the capabilities observed in humans. Partially, this is due to the deployed control paradigms that are mostly based on analytical approaches. The analytical nature of those approaches entails strong model dependencies – regarding the robotic platform as well as the environment – which makes them prone to unknown disturbances. Recently, an increasing number of biologically-inspired control approaches have been presented from which a human-like bipedal gait emerges. Although the control structures only rely on proprioceptive sensory information, the smoothness of the motions and the robustness against external disturbances is impressive. Due to the lack of suitable robotic platforms, until today the controllers have been mostly applied to
simulations.
Therefore, as the first step towards a suitable platform, this thesis presents the Compliant Robotic Leg (CARL) that features mono- as well as biarticular actuation. The design is driven by a set of core-requirements that is primarily derived from the biologically-inspired behavior-based bipedal locomotion control (B4LC) and complemented by further functional aspects from biomechanical research. Throughout the design process, CARL is understood as a unified dynamic system that emerges from the interplay of the mechanics, the electronics, and the control. Thus, having an explicit control approach and the respective gait in mind, the influence of each subsystem on the characteristics of the overall system is considered
carefully.
The result is a planar robotic leg whose three joints are driven by five highly integrated linear SEAs– three mono- and two biarticular actuators – with minimized reflected inertia. The SEAs are encapsulated by FPGA-based embedded nodes that are designed to meet the hard application requirements while enabling the deployment of a full-featured robotic framework. CARL’s foot is implemented using a COTS prosthetic foot; the sensor information is obtained from the deformation of its main structure. Both subsystems are integrated into a leg structure that matches the proportions of a human with a size of 1.7 m.
The functionality of the subsystems, as well as the overall system, is validated experimentally. In particular, the final experiment demonstrates a coordinated walking motion and thereby confirms that CARL can produce the desired behavior – a natural looking, human-like gait is emerging from the interplay of the behavior-based walking control and the mechatronic system. CARL is robust regarding impacts, the redundant actuation system can render the desired joint torques/impedances, and the foot system supports the walking structurally while it provides the necessary sensory information. Considering that there is no movement of the upper trunk, the angle and torque profiles are comparable to the ones found in humans.
Coating of particles is a widely used technique in order to obtain the desired surface modification of the final product, e.g., specific color or taste. Especially in the pharmaceutical industry, rotor granulators are used to produce round, coated pellets. In this work, the coating process in a rotor granulator is investigated numerically using computational fluid dynamics (CFD) coupled with the discrete element method (DEM). The droplets are generated as a second particulate phase in DEM. A liquid bridge model is implemented in the DEM model to take the capillary and viscous forces during the wet contact of the particles into account. A coating model is developed, where the drying of the liquid layer on the particles, as well as the particle growth, is considered. The simulation results of the dry process compared to the simulations with liquid injection show an important influence of the liquid on the particle dynamics. The formation of liquid bridges and the viscous forces in the liquid layer lead to an increase of the average particle velocity and contact time. Changing the injection rate of water has an influence on the contact duration but no significant effect on the particle dynamics. In contrast, the aqueous binder solution has an important influence on the particle movement.
