Hochdruck-Mehrphasengleichgewichte in Systemen aus Wasser, einem organischen Lösungsmittel und (nahekritischem) Ethen

High-pressure multiphase equilibria in systems of water, an organic solvent and („near-critical“) ethene

  • In dieser Arbeit wurden experimentelle und theoretische Untersuchungen zum (nahekritischen) Hochdruck-Mehrphasengleichgewicht ternärer Systeme bestehend aus Ethen, Wasser und einem bei Umgebungsbedingungen vollständig wasserlöslichen organischen Lösungsmittel durchgeführt. Die Untersuchungen behandeln die Grundlagen eines neuartigen Flüssig-flüssig-Extraktionsverfahrens für Naturstoffe. Dieses Extraktionsverfahren wird durch einen wässrig-organischen Flüssigphasensplit ermöglicht, der durch das Aufpressen eines Gases (in der Nähe seines kritischen Zustandes) auf eine homogene wässrig / organische Phase auftritt. Die Untersuchung des durch den Flüssigphasensplit erzeugten Dreiphasengleichgewichts (LLV) sowie der Verteilung ausgewählter Naturstoffe zwischen den beiden Flüssigphasen des Dreiphasengleichgewichts bilden den Schwerpunkt dieser Arbeit. Die Arbeit baut auf Untersuchungen von Wendland (1994) und Adrian (1997) auf. Wendland hat das Phasenverhalten der ternären Systeme Kohlendioxid + Wasser + (Aceton bzw. 2-Propanol) vermessen und umfangreiche Fortran-Routinen zur Beschreibung des ternären Phasenverhaltens sowie der binären Randsysteme entwickelt. Adrian (1997) hat die drei ternären Systeme Kohlendioxid + Wasser + (1- / 2-Propanol bzw. Propionsäure) sowie die Verteilung von zehn organischen Naturstoffen bzw. Modellkomponenten auf die koexistierenden flüssigen Phasen des Dreiphasengleichgewichts LLV in einem (teilweise beiden) der ternären Systeme Kohlendioxid + Wasser + (Aceton bzw. 1-Propanol) untersucht. In dieser Arbeit wurde größtenteils Ethen als nahekritisches Gas eingesetzt, da es im Gegensatz zu dem zuvor benutzten Kohlendioxid in wässrigen Lösungen undissoziiert vorliegt und somit nicht den pH-Wert der koexistierenden flüssigen Phasen bestimmt. Die experimentelle Untersuchung der Phasengleichgewichte erfolgte mit einer Phasengleichgewichtsapparatur, die nach der analytischen Methode arbeitet. In einer thermostatisierten Hochdrucksichtzelle (30 cm3) wurde ein Phasengleichgewicht zwischen mehreren koexistierenden Phasen eingestellt. An die Messzelle waren zwei externe Probenahmeschleifen angeschlossen, durch welche die (in der Zelle koexistierenden) Phasen gepumpt wurden und aus denen Proben für die Analyse mittels GC und HPLC entnommen wurden. Bei Temperaturen zwischen 293 und 333 K und Drücken bis 20.5 MPa wurde das Hochdruck-Mehrphasengleichgewicht (LLV) der beiden ternären Systeme Ethen + Wasser + (1- bzw. 2-Propanol) untersucht. Darüber hinaus wurden die Druck-Temperatur-Koordinaten kritischer Endpunktlinien in diesen beiden ternären Systemen bestimmt und weitere Untersuchungen zum generellen Phasenverhalten angestellt. Den Schwerpunkt dieser Arbeit bildeten Messungen zur Verteilung von Naturstoffen auf die koexistierenden flüssigen Phasen des Dreiphasengleichgewichts LLV: Es wurde u. a. die einzelne Verteilung dreier Paare chemisch ähnlicher Naturstoffe im ternären System Ethen + Wasser + 2-Propanol bei 293 und 333 K untersucht. Die Paare waren 2,5-Hexanediol / 2,5-Hexandion, N-Acetyl-Glukosamin / N-Acetyl-Mannosamin und D- / L-Phenylalanin. Im theoretischen Teil dieser Arbeit wurde das Phasenverhalten der ternären Systeme mit der kubischen Zustandsgleichung (EoS) von Peng und Robinson (1976) in der Modifikation von Melhem et al. (1989) kombiniert mit verschiedenen Mischungsregeln beschrieben. Hierbei wurde sowohl eine Vorhersage des ternären Phasenverhaltens aus Informationen zu den binären Randsystemen als auch die Korrelation des ternären Phasenverhaltens angestrebt. Auch aufgrund der teilweise sehr geringen Anzahl von binären Messpunkten, konnte das Verhalten des Systems Ethen + Wasser + 2-Propanol nicht aus den Informationen zu den binären Randsystemen vorhergesagt werden. Für das Verhalten des ternären Systems Ethen + Wasser + 1-Propanol stimmte die Vorhersage nur qualitativ mit den Messwerten überein. Die Wiedergabe des Phasenverhaltens der ternären Systeme verbesserte sich signifikant, wenn die binären Wechselwirkungsparameter an experimentelle Daten für das Dreiphasengleichgewicht der ternären Systeme angepasst wurden. Zur Korrelation der Verteilung der Naturstoffe auf die flüssigen Phasen des Dreiphasengleichgewichts (LLV) wurde eine Methode benutzt, die auf der Anpassung von Reinstoffparametern der Naturstoffe für die Peng-Robinson EoS basiert. Hierbei wurden die in der EoS benötigten Reinstoffparameter des Naturstoffes an die Ergebnisse der Verteilungsmessungen angepasst, wobei die Wechselwirkungsparameter des ternären Grundsystems übernommen und die Parameter für Wechselwirkungen des Naturstoffes mit dem ternären Grundsystem vernachlässigt wurden. Durch dieses Vorgehen beschreiben die Reinstoffparameter der Naturstoffe auch die Mischungseigenschaften. Durch eine Normierung der Mess- bzw. Rechenwerte auf die gemessenen bzw. berechneten ternären oberen und unteren Begrenzungspunkte des Dreiphasengleichgewichts wurde eine quantitative Beschreibung der Messwerte erhalten.
  • In thermal process engineering there is an increasing number of innovative technologies used for the separation of liquid mixtures. Liquid-liquid extraction is a widely used operation in downstream processing in biotechnology. A successful extraction of natural products demands a non-denaturing environment for the targeted natural product. In the liquid-liquid extraction process, this environment encompasses ambient temperature, non-toxic water-like solvents, specific pH and ionic strength. Due to these special demands on extraction media and a big variety of to be extracted natural products, there is an ongoing search for new water-like extraction media. A promising system consisting of two hydrophilic liquid phases was first described by Elgin and Weinstock (1959). They reported that a mixture of water and a completely water-soluble organic solvent could be forced to split into two water-like liquid phases by pressurisation with a ‘near-critical’ gas (i.e., a substance which at ambient conditions is a gas, near its critical temperature). As a basis for a new mild liquid-liquid extraction process, this phenomenon was first reported by DiAndreth (1985). In recent years, the liquid phase split in ternary systems of ‘near critical’ carbon dioxide + water + hydrophilic organic solvent has been investigated intensively (Adrian et al., 1998). In addition to the basic phase equilibrium phenomenon, partitioning of natural products into coexisting water-like liquid phases has been studied. This contribution extends existing experimental knowledge and modelling methods on the basic phase split phenomenon and the partitioning of natural products into coexisting water-like liquid phases. Therefore, systems comprising of near-critical ethene were investigated. As the usage of ethene enables the adjustment of the pH of the coexisting liquid phases, being in contrast to the usage of carbon dioxide. The experimental investigations were carried out in a measurement apparatus based on the analytical method. The central part of the equipment was a cylindrical, rotating, high-pressure view cell with an internal volume of about 30 cm3. The cell was filled by means of high-pressure hand pumps with the liquids. A liquid-liquid phase split could be achieved by pressurisation of the homogeneous liquid with the near critical gas. For analysis two of three coexisting fluid phases were pumped through two separate, external sample loops. Special sample valves were incorporated into these loops to allow for online analysis by GC and HPLC. Two ternary ethene containing systems ethene + water + 1- / 2-propanol were investigated at temperatures ranging from 293 to 333 K and pressures up to 20.5 MPa. The main focus during these investigations was the determination of the composition of the coexisting fluid phases in the liquid-liquid-vapour equilibrium (LLV), and the determination of pressure-temperature co-ordinates of critical endpoint lines. The centre of interest of this work was the experimental investigation of the single partitioning of pairs of natural products – near infinite dilution - into the coexisting liquid phases of the three-phase equilibrium LLV: The partitioning of each member of three pairs (2,5-hexanediol / 2,5-hexandione, N-acetyl-glucosamine / N-acetyl-mannosamine, L- / D-phenylalanine) of chemically similar natural products into the coexisting liquid phases in the ternary system ethene + water + 2-propanol resulted in pairs of very similar partition coefficients (around one or much less). This does not allow for a separation process for the pairs. However, a process of an extraction out of an organic liquid phase or a purification of an aqueous liquid containing these natural products is feasible. In the modelling section of this work, the cubic equation of state according to Peng and Robinson (1976) was used to predict the phase equilibrium of the ternary ethene + water + propanol systems. Several mixing rules were tested and the necessary parameters were fitted to phase equilibrium data of the corresponding binary mixtures. Following the latter procedure, the ternary systems ethene + water + propanol could only be predicted qualitatively from the binary data alone. A dramatic qualitative as well as quantitative improvement in the description of the measured ternary three-phase equilibrium was achieved, when it was exclusively correlated with the measured ternary LLV-data. For the correlation of the natural product the natural product pure component parameters of the Peng-Robinson EoS were correlated with the partitioning results, whereby the interaction parameters of the natural product with the ternary phase building components were set to zero. Applying different standardizations of the measured and correlated pressures yielded good correlation results and allowed even for a nearly quantitative transfer of natural product partition results from one ternary phase-building system to another.
