Auswahl und Verarbeitung von Früchten zur Steigerung der Gehalte an phenolischen Antioxidantien in Fruchtsäften

Selection and processing of fruits to increase the content of polyphenols in fruit juices

  • Da Polyphenole als gesund angesehen werden, ist es Ziel dieser Arbeit, ihre Gehalte in Fruchtsäften zu erhöhen. Dies beinhaltet zum einen das Auffinden polyphenolreicher Apfel- und Beerenobstsorten als geeignete Rohware. Gleichzeitig entsteht dabei ein Datensatz über sortenreine Apfel- und Beerenobstsäfte, der die RSK-Werte ergänzt. Zum anderen sind Wege zur Minimierung von Verarbeitungsverlusten durch gezielte Studien zur Qualitätssteigerung des Endproduktes Fruchtsaft wichtig. Die im Screening untersuchten sortenreinen Mostapfelsäfte aus drei Jahrgängen zeigen sehr hohe Gesamtphenolgehalte (GP) und antioxidative Kapazitäten (aK), die die Gehalte von Tafeläpfeln übersteigen. Sorten wie Bittenfelder und Weißer Trierer Weinapfel erreichen aK von Rotwein. Beerenobstsäfte sind reicher an Antioxidantien als Apfelsäfte. Innerhalb der Arten konnten besonders antioxidantienreiche Sorten gefunden werden. Bezogen auf die aK lautet die Reihenfolge: Tafelapfel < Mostapfel <= Erdbeere < Himbeere = Brombeere < Cranberry < Heidelbeere < Johannisbeere = Boysenberry < Aronia. Darüber hinaus sind erfolgreich Extraktions- und Analysemethoden zur Bestimmung der verschiedenen Formen von Ellagsäure entwickelt und zur Untersuchung von Erdbeeren und Himbeeren eingesetzt worden. Die Gesamtellagsäuregehalte von Himbeeren übersteigen bisher beschriebene Gehalte deutlich. Darüber hinaus sind Äpfel in die Gewebezonen aufgeteilt und auf Antioxidantien untersucht worden. Dies hat ergeben, dass die Quercetine (Q) fast ausschließlich in der Schale vorhanden sind und die Dihydrochalkone (DHC) größtenteils im Kerngehäuse. Die Phenolcarbonsäuren (PC) kommen ebenso wie die Flavanole (F) in allen Gewebezonen vor. Gerade die schlecht wasserlöslichen DHC und Q, die an den festen Bestandteilen der Frucht sitzen, gehen schlecht in den Saft über. Im Rahmen der Apfelverarbeitungsstudien sind Probleme bei der Probenahme und Extraktion erkannt und behoben worden. Die durchgeführten Verarbeitungsstudien haben ergeben, dass Prozesse zur Erhöhung des Transfers von DHC und Q wie etwa eine längere Maischestandzeit zu einem Verlust von F und PC führen. Dagegen verhindern Maßnahmen zum Schutz vor Oxidation, wie eine zusätzliche KZE des Saftes nach dem Pressen, die Extraktion der DHC und Q. Eine Steigerung des Polyphenolgehaltes kann jedoch durch eine Nachextraktion erreicht werden, wobei die Supratonmaschine keinen Vorteil bringt. Dieser Nachextraktsaft von polyphenolreichen Sorten kann darüber hinaus zur Qualitätssteigerung von einfacheren (Tafel)Apfelsäften eingesetzt werden. Die besten Ergebnisse des Übergangs der Polyphenole von der Frucht in das Getränk konnten bei der Herstellung eines Ganzfruchtproduktes erzielt werden. Selbst nach Verdünnung auf Nektarstärke sind mehr Polyphenole im Getränk enthalten als in einem vergleichbaren Saft. Lagerungsversuche über ein Jahr hinweg zeigen, dass sich Bohnapfelsaft und Mehrfruchtsaft sehr unterschiedlich verhalten. Während der untersuchte Bohnapfelsaft sich im Bezug auf Antioxidantien kaum über die Lagerzeit verändert, nehmen die Anthocyane des Mehrfruchtsaftes schon im ersten Lagermonat deutlich ab. Dagegen verschlechtert sich der Bohnapfelsaft sensorisch schnell während der Mehrfruchtsaft noch nach einem Jahr geschmeckt hat. Dies zeigt die Wichtigkeit sensorischer Untersuchungen bei solchen Studien. Aus polyphenolreichen Säften hergestellte Mehrfruchtsäfte (100% Saft) können als „Wellnessgetränke“ angesehen werden, da sie einen hohen gesundheitlichen Nutzen haben. Neue Rezepturen mit phenolreichen Ausgangssäften und optimierter Verarbeitung sollten weiter entwickelt werden.
  • Polyphenols are important secondary plant metabolites which are considered beneficial to health. Aim of this study was examine how to increase the content of polyphenols in fruit juices. The most appropriate cultivars of apples and berries rich in polyphenols had to be found as raw material for juice processing. Additionally, a way to minimise the decrease of polyphenols during processing was developed in further studies. The analyzed cider apple juices of three harvesting years show very high total phenolic contents and total antioxidant capacities. They exceed the amounts of table apple juices by far. Cultivars like Bittenfelder or Weißer Trierer Weinapfel reach antioxidant capacities of red wine. Berry-juices contain more antioxidants than apple juices. During two years screening, berry varieties rich in polyphenols were found. Fruits can be arranged by their antioxidant capacity as follows: Table apple < cider apple <= strawberry < raspberry = blackberry < cranberry < blueberry < black currant = boysenberry < chokeberry. Additionally, new analytical methods for the determination of free ellagic acid, its glycosides and total ellagic acids (after hydrolysis) have been developed and used to analyze strawberries and raspberries. Total ellagic acid content in raspberries was far more than reported until now. Another aim of this study was the examination of the antioxidants in different parts (peel, parenchyma and core zone) of the apples. The examination has shown that flavonols (quercetins) were nearly exclusively localized in the peel zone and dihydrochalcones above all in the core zone. Phenolic acids and flavanoles have been found in all compartments, phenolic acids especially in the core zone and flavanols mainly in the peel and core zone. The transfer of dihydrochalcones and quercetins into the juice is poor (0-14 % of the quercitines and 14-72 % of the dihydrochalcones get into the juices). Problems concerning sample taking and extraction during processing studies have been revealed and eliminated. Processing studies have shown that processes intended to increase the transfer of dihyrochalcones and quercetines, like increased extraction time of the mash, lead to a reduction of flavanols and phenolic acids in juice. On the other hand, steps that protect polyphenols from oxidation like an additional short term heating step after pressing reduce the extraction of dihydrochalcones and quercetines. But an enhancement of the polyphenol content can be achieved by an additional extraction step of the juice with no advantage of using a supraton machine. Additionally, the extraction juices of varieties rich in polyphenols can be used to improve the quality of simple (table)apple juices. The best results of transfer of polyphenolics into juice could be reached with the production of a whole-fruit-product. Diluted to nectar to make it drinkable, this product contains more polyphenols than a 100%- juice of the same apples. A storage-experiment has been performed with an apple juice and a juice from different fruits. While the Bohnapfel-juice almost does not change analytically during the storage period of over one year, the anthocyanins of a juice from different fruits (“Mehrfruchtsaft”) decrease even during the first month of storage. Whereas the Bohnapfel-juice gets less sensory quality (appearance, odour, taste) after half a year and was refused after one year, the “Mehrfruchtsaft” still tasted good. Thus, sensory aspects shall be considered along with the chemical parameters with the aim to achieve best-quality beverages. Mixtures (100% juice) of juices rich in polyphenols could be seen as wellness-beverages, because of their high value to health. New mixtures with juices rich in polyphenols and optimal processing need further investigations.
